Special Handling Step 1: Prepare esterase inhibitor solution (Neostigmine 2 mg/mL): Weigh 68 mg of Neostigmine Bromide (Sigma Chemical, St. Louis, MO) and transfer to a 25 mL volumetric flask. Dissolve and dilute to volume with deionized water, mix thoroughly. Store in an amber bottle at room temperature and discard nine (9) months after preparation. STEP 2: Prepare collection tubes to 3 mL plasma draw tubes containing Na2EDTA anti-coagulant, add 20 microliters of the 2 mg/mL Neostigmine solution without breaking the vacuum. This is accomplished by using a 25 microliter syringe and piercing the rubber top of the container. STEP 3: Collecting samples and storage: Draw blood into the prepared collection tubes, making efforts to draw at least 2.5 mL. Invert tube at least eight (8) times to mix with the EDTA and Neostigmine. Immediately centrifuge and carefully transfer, with plastic pipette, the supernatant plasma into a labeled plastic container. Keep refrigerated and freeze if stored for more than one week.
